Nitrate is the major form of macronutrient nitrogen that plants take up from the soil. Fluctuating availability or deficiency of this element severely limits plant growth and negatively affects crop production in the agricultural system. To cope with the heterogeneity of nitrate distribution in soil, plants evolved a complex regulatory mechanism that allows rapid adjustment of physiological and developmental processes to the status of this nutrient. The root, as a major exploitation organ that controls the uptake of nitrate to the plant body, acts as a regulatory hub that, according to nitrate availability, coordinates the growth and development of other plant organs. Here, we identified a regulatory framework, where cytokinin response factors (CRFs) play a central role as a molecular readout of the nitrate status in roots to guide shoot adaptive developmental response. We show that nitrate-driven activation of NLP7, a master regulator of nitrate response in plants, fine tunes biosynthesis of cytokinin in roots and its translocation to shoots where it enhances expression of CRFs. CRFs, through direct transcriptional regulation of PIN auxin transporters, promote the flow of auxin and thereby stimulate the development of shoot organs.","lang":"eng"}],"volume":119,"acknowledgement":"We acknowledge Hana Semeradova, Juan Carlos Montesinos, Nicola Cavallari, Marc¸al Gallem\u0003ı, Kaori Tabata, Andrej Hurn\u0003y, and Sascha Waidmann for sharing materials; and Marina Borges Osorio for critical reading of the manuscript. Plants have evolved different strategies for sensing and responding to heterogeneous nitrogen distribution. Modulation of root system architecture, including primary root growth and branching, is among the most essential plant adaptions to ensure adequate nitrogen acquisition. However, the immediate molecular pathways coordinating the adjustment of root growth in response to distinct nitrogen sources, such as nitrate or ammonium, are poorly understood. Here, we show that growth as manifested by cell division and elongation is synchronized by coordinated auxin flux between two adjacent outer tissue layers of the root. This coordination is achieved by nitrate‐dependent dephosphorylation of the PIN2 auxin efflux carrier at a previously uncharacterized phosphorylation site, leading to subsequent PIN2 lateralization and thereby regulating auxin flow between adjacent tissues. A dynamic computer model based on our experimental data successfully recapitulates experimental observations. Our study provides mechanistic insights broadening our understanding of root growth mechanisms in dynamic environments."}],"date_updated":"2024-03-25T23:30:22Z","year":"2021","citation":{"ama":"Ötvös K, Marconi M, Vega A, et al. Local fluctuations in water and nutrient availability, sudden changes in temperature or other abiotic and biotic stresses can trigger changes in the growth of plant organs. Multiple mutually interconnected hormonal signaling cascades act as essential endogenous translators of these exogenous signals in the adaptive responses of plants. Although the molecular backbones of hormone transduction pathways have been identified, the mechanisms underlying their interactions are largely unknown. Here, using genome wide transcriptome profiling we identify an auxin and cytokinin cross-talk component; SYNERGISTIC ON AUXIN AND CYTOKININ 1 (SYAC1), whose expression in roots is strictly dependent on both of these hormonal pathways. We show that SYAC1 is a regulator of secretory pathway, whose enhanced activity interferes with deposition of cell wall components and can fine-tune organ growth and sensitivity to soil pathogens.","lang":"eng"}],"ec_funded":1,"quality_controlled":"1","file_date_updated":"2020-10-06T07:47:53Z","publisher":"Springer Nature","article_type":"original","scopus_import":"1","_id":"7805","pmid":1,"author":[{"id":"4DC4AF46-F248-11E8-B48F-1D18A9856A87","last_name":"Hurny","first_name":"Andrej","full_name":"Hurny, Andrej","orcid":"0000-0003-3638-1426"},{"id":"33A3C818-F248-11E8-B48F-1D18A9856A87","first_name":"Candela","last_name":"Cuesta","orcid":"0000-0003-1923-2410","full_name":"Cuesta, Candela"},{"first_name":"Nicola","last_name":"Cavallari","full_name":"Cavallari, Nicola","id":"457160E6-F248-11E8-B48F-1D18A9856A87"},{"last_name":"Ötvös","first_name":"Krisztina","full_name":"Ötvös, Krisztina","orcid":"0000-0002-5503-4983","id":"29B901B0-F248-11E8-B48F-1D18A9856A87"},{"full_name":"Duclercq, Jerome","first_name":"Jerome","last_name":"Duclercq"},{"first_name":"Ladislav","last_name":"Dokládal","full_name":"Dokládal, Ladislav"},{"id":"310A8E3E-F248-11E8-B48F-1D18A9856A87","full_name":"Montesinos López, Juan C","orcid":"0000-0001-9179-6099","last_name":"Montesinos López","first_name":"Juan C"},{"id":"460C6802-F248-11E8-B48F-1D18A9856A87","last_name":"Gallemi","first_name":"Marçal","full_name":"Gallemi, Marçal","orcid":"0000-0003-4675-6893"},{"id":"42FE702E-F248-11E8-B48F-1D18A9856A87","last_name":"Semeradova","first_name":"Hana","full_name":"Semeradova, Hana"},{"full_name":"Rauter, Thomas","first_name":"Thomas","last_name":"Rauter","id":"A0385D1A-9376-11EA-A47D-9862C5E3AB22"},{"full_name":"Stenzel, Irene","last_name":"Stenzel","first_name":"Irene"},{"full_name":"Persiau, Geert","last_name":"Persiau","first_name":"Geert"},{"full_name":"Benade, Freia","first_name":"Freia","last_name":"Benade"},{"full_name":"Bhalearo, Rishikesh","last_name":"Bhalearo","first_name":"Rishikesh"},{"full_name":"Sýkorová, Eva","first_name":"Eva","last_name":"Sýkorová"},{"full_name":"Gorzsás, András","last_name":"Gorzsás","first_name":"András"},{"last_name":"Sechet","first_name":"Julien","full_name":"Sechet, Julien"},{"first_name":"Gregory","last_name":"Mouille","full_name":"Mouille, Gregory"},{"full_name":"Heilmann, Ingo","first_name":"Ingo","last_name":"Heilmann"},{"first_name":"Geert","last_name":"De Jaeger","full_name":"De Jaeger, Geert"},{"full_name":"Ludwig-Müller, Jutta","last_name":"Ludwig-Müller","first_name":"Jutta"},{"id":"38F4F166-F248-11E8-B48F-1D18A9856A87","orcid":"0000-0002-8510-9739","full_name":"Benková, Eva","first_name":"Eva","last_name":"Benková"}],"date_created":"2020-05-10T22:00:48Z","article_processing_charge":"No","department":[{"_id":"EvBe"}],"publication_status":"published","intvolume":" 11","title":"Synergistic on Auxin and Cytokinin 1 positively regulates growth and attenuates soil pathogen resistance"},{"day":"01","doi":"10.15252/embj.2019104238","abstract":[{"text":"Cell production and differentiation for the acquisition of specific functions are key features of living systems. The dynamic network of cellular microtubules provides the necessary platform to accommodate processes associated with the transition of cells through the individual phases of cytogenesis. Here, we show that the plant hormone cytokinin fine‐tunes the activity of the microtubular cytoskeleton during cell differentiation and counteracts microtubular rearrangements driven by the hormone auxin. The endogenous upward gradient of cytokinin activity along the longitudinal growth axis in Arabidopsis thaliana roots correlates with robust rearrangements of the microtubule cytoskeleton in epidermal cells progressing from the proliferative to the differentiation stage. Controlled increases in cytokinin activity result in premature re‐organization of the microtubule network from transversal to an oblique disposition in cells prior to their differentiation, whereas attenuated hormone perception delays cytoskeleton conversion into a configuration typical for differentiated cells. Intriguingly, cytokinin can interfere with microtubules also in animal cells, such as leukocytes, suggesting that a cytokinin‐sensitive control pathway for the microtubular cytoskeleton may be at least partially conserved between plant and animal cells.","lang":"eng"}],"year":"2020","citation":{"apa":"Montesinos López, J. The Arabidopsis root is a widely used model for developmental, physiological and cell biological studies. Live imaging greatly aids these efforts, but the horizontal sample position and continuous root tip displacement present significant difficulties. Here, we develop a confocal microscope setup for vertical sample mounting and integrated directional illumination. We present TipTracker – a custom software for automatic tracking of diverse moving objects usable on various microscope setups. Combined, this enables observation of root tips growing along the natural gravity vector over prolonged periods of time, as well as the ability to induce rapid gravity or light stimulation. We also track migrating cells in the developing zebrafish embryo, demonstrating the utility of this system in the acquisition of high-resolution data sets of dynamic samples. The architecture of the root system is controlled by endogenous mechanisms that constantly integrate environmental signals, such as availability of nutrients and water. Extremely important for efficient soil exploitation and survival under less favorable conditions is the developmental flexibility of the root system that is largely determined by its postembryonic branching capacity. Modulation of initiation and outgrowth of lateral roots provides roots with an exceptional plasticity, allows optimal adjustment to underground heterogeneity, and enables effective soil exploitation and use of resources. Here we discuss recent advances in understanding the molecular mechanisms that shape the plant root system and integrate external cues to adapt to the changing environment."}],"citation":{"ista":"Ötvös K, Benková E. 2017. Although cytokinin-mediated modulation of auxin distribution is a developmentally crucial hormonal interaction, its molecular basis is largely unknown. Here we show a direct regulatory link between cytokinin signalling and the auxin transport machinery uncovering a mechanistic framework for cytokinin-auxin cross-talk. We show that the CYTOKININ RESPONSE FACTORS (CRFs), transcription factors downstream of cytokinin perception, transcriptionally control genes encoding PIN-FORMED (PIN) auxin transporters at a specific PIN CYTOKININ RESPONSE ELEMENT (PCRE) domain. Removal of this cis-regulatory element effectively uncouples PIN transcription from the CRF-mediated cytokinin regulation and attenuates plant cytokinin sensitivity. We propose that CRFs represent a missing cross-talk component that fine-tunes auxin transport capacity downstream of cytokinin signalling to control plant development.","lang":"eng"}],"day":"01","doi":"10.1038/ncomms9717","citation":{"short":"M.
